Skeletal muscle as an experimental model of choice to study tissue aging and rejuvenation.
Skeletal muscle is among the most age-sensitive tissues in mammal organisms. Significant changes in its resident stem cells (i.e., satellite cells, SCs), differentiated cells (i.e., myofibers), and extracellular matrix cause a decline in tissue homeostasis, function, and regenerative capacity. Based on the conservation of aging across tissues and taking advantage of the relatively well-characterization of the myofibers and associated SCs, skeletal muscle emerged as an experimental system to study the decline in function and maintenance of old tissues and to explore rejuvenation strategies. In this review, we summarize the approaches for understanding the aging process and for assaying the success of rejuvenation that use skeletal muscle as the experimental system of choice. We further discuss (and exemplify with studies of skeletal muscle) how conflicting results might be due to variations in the techniques of stem cell isolation, differences in the assays of functional rejuvenation, or deciding on the numbers of replicates and experimental cohorts
Ipteks Balance Scorecard di Dinas Komunikasi Informatika Persandian dan Statistik Provinsi Sulawesi Utara
Performance is the achievement or level of success of a person or group of people who have exceeded the standard set. The higher the achievement of the work the better it will be and vice versa. To find out the achievement, it is necessary to measure performance. In the scope of the public sector where performance measurement is very important because performance measurement can help to determine the level of achievement of the success of activities or work program from an agency both within the scope of the region to the center. For the reason, in knowing the level of performance it is necessary to measure performance. Performance measurement in this study uses a balanced scorecard concept, where the performance measurement is assessed based on four perspectives. Where from these four perspectives will be seen the performance achieved by the Office of Communication, Information, Coding and Statistics of North Sulawesi Province. Through the four perspectives of the balance scorecard it will be known in any scope the performance of the Regional Office of Communication, Information, Coding and Statistics of North Sulawesi Province still needs to be emproved
Gambaran Status Gizi Atlet Angkat Berat, Angkat Besi Dan Binaraga Komite Olahraga Nasional Indonesia (Koni) Provinsi Riau Tahun 2015
A great nation can be recognized as a great nation if they haveextraordinary achievement and perfomance on International sport event.Nowadays, there are crisis of sports in Indonesia due to lack of Internationalsport achievements and do not show a lot of improvements in national sportdevelopment. Achievement on sports influenced by some factors, one wasbalanced nutrition on athletes. Balanced nutrition on athletes should be givenhuge attention, because giving unbalanced nutrition will cause damage toathletes\u27 performance, health and sport achievement. Theresearch design is descriptive cross-sectional. The samples were obtained by totalsampling. The result showed that there were 30 people : 16 heavy lifters, 9weightlifters and 5 bodybuilder; 70% male and 30% female; the average age is atthe end of adolescent age (17-25 yr); Body Mass Index (BMI) mostly overweight(33,3%); the measurement of waist circumference showed that 43,3% has obesity;waist hip ratio showed that 46,6% has obesity, body fat compotition by skinfoldshow abnormality by 63,3% and bioelectrical impedance analysis (BIA) by 90%.The conclusion is athletes\u27 nutrition status is high normal. Based on the researchKONI suggested to show more attention to athletes\u27 nutrition
Pengaruh Kompetensi Sumber Daya Manusia, Teknologi Informasi, Reviu Laporan Keuangan dan Sistem Pengendalian Internal terhadap Kualitas Laporan Keuangan pada Pemerintah Provinsi Sulawesi Utara
This study aims to examine the effect of human resources competence, information technology, review of financial statement and internal control system on the quality of financial statement in Government of North Sulawesi. The variables in this study consist of independent variable :human resources competence (X1), information technology (X2), review of financial statement (X3) and internal control system (X4).On the other hand, the dependent variable is the quality of financial statement (Y). Data were analyzed by multiple linear regression analysis (multiple regression). The results indicate that human resources competence, information technology, review of financial statement and internal control system simultaneously have a significant influence on the quality of financial statement. The value of the coefficient of determination shows that human resources competence, information technology, review of financial statement and internal control system simultaneously contribute to the quality of financial statement.R2 is 71.3%, it means those dependent variables affect the quality of financial statement by 71.3%. While the remaining of 28.7% is influenced by other variables which are excluded within this research. Partial test results show that information technology, review of financial statement and internal control system have a significant impact on the quality of financial statement. Human resources competence has no significant impact on the quality of financial statement. Keywords : quality of financial statement, human resources competence, information technology, review of financial statement, internal control syste
Young stellar population and ongoing star formation in the HII complex Sh2-252
In this paper an extensive survey of the star forming complex Sh2-252 has
been undertaken with an aim to explore its hidden young stellar population as
well as to understand the structure and star formation history. This complex is
composed of five embedded clusters associated with the sub-regions A, C, E, NGC
2175s and Teu 136. Using 2MASS-NIR and Spitzer-IRAC, MIPS photometry we
identified 577 young stellar objects (YSOs), of which, 163 are Class I, 400 are
Class II and 14 are transition disk YSOs. Spatial distribution of the candidate
YSOs shows that they are mostly clustered around the sub-regions in the western
half of the complex, suggesting enhanced star formation activity towards its
west. Using the spectral energy distribution and optical colour-magnitude
diagram based age analyses, we derived probable evolutionary status of the
sub-regions of Sh2-252. Our analysis shows that the region A is the youngest (~
0.5 Myr), the regions B, C and E are of similar evolutionary stage (~ 1-2 Myr)
and the clusters NGC 2175s and Teu 136 are slightly evolved (~ 2-3 Myr).
Morphology of the region in the 1.1 mm map shows a semi-circular shaped
molecular shell composed of several clumps and YSOs bordering the western
ionization front of Sh2-252. Our analyses suggest that next generation star
formation is currently under way along this border and that possibly
fragmentation of the matter collected during the expansion of the HII region as
one of the major processes responsible for such stars. We observed the densest
concentration of YSOs (mostly Class I, ~ 0.5 Myr) at the western outskirts of
the complex, within a molecular clump associated with water and methanol masers
and we suggest that it is indeed a site of cluster formation at a very early
evolutionary stage, sandwiched between the two relatively evolved CHII regions

Rejuvenation of brain, liver and muscle by simultaneous pharmacological modulation of two signaling determinants, that change in opposite directions with age.
We hypothesize that altered intensities of a few morphogenic pathways account for most/all the phenotypes of aging. Investigating this has revealed a novel approach to rejuvenate multiple mammalian tissues by defined pharmacology. Specifically, we pursued the simultaneous youthful in vivo calibration of two determinants: TGF-beta which activates ALK5/pSmad 2,3 and goes up with age, and oxytocin (OT) which activates MAPK and diminishes with age. The dose of Alk5 inhibitor (Alk5i) was reduced by 10-fold and the duration of treatment was shortened (to minimize overt skewing of cell-signaling pathways), yet the positive outcomes were broadened, as compared with our previous studies. Alk5i plus OT quickly and robustly enhanced neurogenesis, reduced neuro-inflammation, improved cognitive performance, and rejuvenated livers and muscle in old mice. Interestingly, the combination also diminished the numbers of cells that express the CDK inhibitor and marker of senescence p16 in vivo. Summarily, simultaneously re-normalizing two pathways that change with age in opposite ways (up vs. down) synergistically reverses multiple symptoms of aging
Investigation into the role of the germline epigenome in the transmission of glucocorticoid-programmed effects across generations.
BACKGROUND: Early life exposure to adverse environments affects cardiovascular and metabolic systems in the offspring. These programmed effects are transmissible to a second generation through both male and female lines, suggesting germline transmission. We have previously shown that prenatal overexposure to the synthetic glucocorticoid dexamethasone (Dex) in rats reduces birth weight in the first generation (F1), a phenotype which is transmitted to a second generation (F2), particularly through the male line. We hypothesize that Dex exposure affects developing germ cells, resulting in transmissible alterations in DNA methylation, histone marks and/or small RNA in the male germline. RESULTS: We profile epigenetic marks in sperm from F1 Sprague Dawley rats expressing a germ cell-specific GFP transgene following Dex or vehicle treatment of the mothers, using methylated DNA immunoprecipitation sequencing, small RNA sequencing and chromatin immunoprecipitation sequencing for H3K4me3, H3K4me1, H3K27me3 and H3K9me3. Although effects on birth weight are transmitted to the F2 generation through the male line, no differences in DNA methylation, histone modifications or small RNA were detected between germ cells and sperm from Dex-exposed animals and controls. CONCLUSIONS: Although the phenotype is transmitted to a second generation, we are unable to detect specific changes in DNA methylation, common histone modifications or small RNA profiles in sperm. Dex exposure is associated with more variable 5mC levels, particularly at non-promoter loci. Although this could be one mechanism contributing to the observed phenotype, other germline epigenetic modifications or non-epigenetic mechanisms may be responsible for the transmission of programmed effects across generations in this model
